No, it hasn't. You still have the freedom to speak your mind. You can simply use another means to do so.

YouTube, Facebook, and Twitter are not God. There are plenty of forums, video sites, facebook like site, and twitter like sites that you can use.
You could start your own blog, set up your own radio station, even build your own forum, video site, facebook site, or twitter like site.

The 1st amendment does not apply to anything but Congress. It only applies to the federal government. It prohibits Congress from passing certain laws.
